The Inflammatory Response

Inflammation is a natural and protective reaction in response to tissue damage, cellular injury, pathogen invasion or irritants.

The goal of inflammation is to recruit phagocytes and plasma proteins to the injured or invaded area in order to:

  1. Isolate, destroy or inactivate invaders (phagocytosis)
  2. Remove cell debris and clean the area (efferocytosis)
  3. Prepare the subsequent healing and repair

Inflammation is a self-limited process that enables healing and a return to homeostasis.


INITIATION Phase – Driven by proinflammatory mediators – Leukotrienes and Prostaglandins.
During this phase, leucocytes and PMN neutrophils are recruited to the damaged tissue where PMN phagocyte the foreign particles and tissue debris. At this time, heat, redness, swelling and pain take part.

RESOLUTION Phase – Driven by pro-resolving mediators – SPMs.
Specialized Pro-resolving Mediators are essential to end the process by promoting tissue repair and regeneration. In this way, they are responsible for the termination of PMN recruitment, the counter regulation of pro-inflammatory mediators, efferocytosis (debris clearance) and tissue remodeling.

At one point during the inflammatory process, a switch in lipid mediators occurs which induces the resolution process. Lipoxins, specifically Lipoxin A4, are key lipid mediators critical to resolution because they signal the end of PMN recruitment and enable biosynthesis of SPMs: resolvins, protectins and maresins.
